Test Principal:
Malaria P.f Antigen Rapid Test is a lateral flow immunochromatographic assay for the qualitative differential detection of P. falciparum specific histidine rich protein-2 (Pf. HRP-2) in human whole blood. The test utilizes colloidal gold conjugated monoclonal anti-P. falciparum HRP-2 antibodies and monoclonal anti-P. falciparum HRP-2 antibodies immobilized on a nitrocellulose membrane at the test line (P.f region). The blood sample is added to the sample well, followed by the addition of assay buffer. The buffer lyses the red blood cells and facilitates the migration of the sample through the membrane by capillary action. If P. falciparum HRP-2 antigen is present in the specimen, it binds with the colloidal gold-labelled monoclonal antibodies to form an antigen-antibody complex.
This complex migrates along the membrane and is captured by the immobilized monoclonal antibodies at the test line region, producing a colored band, indicating a reactive result (P.f Positive). If the specimen does not contain P. falciparum HRP-2 antigen, no colored band appears in the test region, indicating a non-reactive result. A colored procedural control line appearing at the “C” region confirms proper sample migration and correct performance of the test procedure.
